ASSAY PRINCIPLE:
Diazyme Aβ40 CLIA Kit is used in combination with Diazyme Aβ42 (Catalog# DZ191A) for the quantitative determination of Aβ40 in plasma. Diazyme Aβ40 CLIA Kit is a sandwich chemiluminescent immunoassay that uses magnetic microbeads coated with an anti-Aβ40 monoclonal antibody for capture, and an anti-Amyloid beta monoclonal antibody labeled with ALP for detection.
Sample or calibrator or control is mixed with the capture reagent and the mixture is incubated at 37°C to form a complex. After sedimentation in a magnetic field, the antibody-Aβ40 complex is washed to remove unbound proteins and substances. Subsequently, the anti-Amyloid beta monoclonal antibody labeled with ALP is added and all components are incubated again to form a sandwich complex. After washing the final complex, the substrate reagents are added, and a flash chemiluminescent reaction is initiated. The light signal is measured by a photomultiplier as RLU, and the signal is proportional to the concentration of Aβ40 present in the samples.
