| Product Features The assay provides an extended linear range up to 250 U/L with virtually no interferences from triglyceride (300 mg/dL), hemoglobin (150 mg/dL), and bilirubin up to (20 mg/dL). The Diazyme Lipase assay has high precision with Inter-Run %CV’s of less than 5%, and Intra-Run %CV’s of less than 6% at 215 U/L. The assay has an excellent correlation to Beckman™ Lipase reagent with r2 = 0.997. Reagents are stable for 12 months from date of manufacture when stored at 2°-8°C. |

Assay PrincipleThe method for the determination of lipase is based on the cleavage of specific chromogenic lipase substrate 1,2-O-dilaurylrac-glycero-3-glutaric acid-(6'methyl-resorufin)-ester emulsified in stabilized micro-particles. In the presence of specific activators of pancreatic lipase as colipase, calcium ions and bile acids, the substrate is converted to 1,2-O-dilauryl-rac-glycerol and glutaric acid-6'-methylresorufinester which decomposes spontaneously to glutaric acid and methylresorufin. The increase of absorbance at 580 nm, due to methylresorufin formation, is proportional to the activity of lipase in the sample. |

| Intended use The Diazyme Lipase assay in conjunction with lipase assay calibrator set is intended for the in vitro quantitative determination of lipase in serum or plasma. |
