The assay shows virtually no affect by the following interfering substances at indicated concentrations: Na+ 200 mM, NH4+ 0.5 mM, Ca2+ 5.0 mM, Mg2+ 5.0 mM, ascorbic acid 5.0 mM, and bilirubin 10 mg/dl, 1.5 mM Pi, 0.25 mM CuCl2, and 5 mM glucose. The assay has a linearity range from 2-8 mM. Within precision %CVs are 3.2% and 3.0% for 4.4 mM and 6.4 mM potas-sium serum samples, respectively. Total precision %CVs are 5.3% and 3.3% for 4.4 mM and 6.4 mM potassium serum samples, respectively. Comparison study showed that the assay has good correlation with existing ISE method with a correlation coefficient of 0.97. Reconstituted reagents are stable for 2 weeks @ 2°-8° C.
Assay Principle
Potassium is determined spectrophotometrically through a kinetic coupling assay system using potassium dependent S-Adenosylmethionine Synthase (MAT). NADH is generated after a series of enzymatic reactions. The corresponding increase of optical density at 340nm is proportional to the potassium concentration in the serum.

For the quantitative in vitro determination of Potassium in serum samples.
