Product Features
The assay is ready-to-use for manual methods and adaptable for analyzers. The assay is specific for NAG and has no detectable reaction with other glycosidases and has excellent sensitivity with a linear range from up to 200 U/L and excellent precision (CV <5%) . There is no significant interference from hemoglobin or albumin. Interference from bilirubin occurs only at levels higher than 5 mg/dL. The reagent is stable for 12 months when stored at @ 2°-8°C.
Assay Principle
The Diazyme NAG assay uses a specific substrate, 2-methoxy-4-(2’nitrovinyl)-phenyl 2-acetamido-2-deoxy-β-D-glucopyranoside (MNP-GlcNAc) that is hydrolyzed by NAG to produce 2-methoxy-4-(2’-nitrovinyl)-phenol product. The product formation is detected at 505 nm upon addition of an alkaline buffer.
Intended use
The N-acetyl-β-D-glucosaminidase (NAG) assay kit is for the quantitative in vitro determination of NAG activity in serum or urine samples. Increased NAG levels in urine are an early indication of renal disease and can serve as a valuable renal monitoring test in disorders such as nephritic syndrome, glomerulunephritis, drug abuse associated nephrotoxicity, diabetes-associated nephropathy, hypertension and urinary tract infections. In the USA, the assay is for research use only.
