| Product Features The Diazyme Total Bile Acids assay is a liquid stable system, ready to use for both manual methods and adaptable for many automated chemistry analyzers. The assay has excellent sensitivity with a linear range from 1-180 µmole/L and has been validated for both human and veterinary applications. Diazyme’s unique enzyme cycling method amplifies signal for accurate and fast measurement using a small sample up to 50% less than conventional NBT methods making it an excellent choice for both veterinary and pediatric applications. The assay has excellent precision (%CV <5%) with no significant interference from lipemic or hemolytic samples. Packaged for optimal operator convenience reagent transfer can be eliminated for most chemistry systems with instrument specific packaging options including Roche Hitachi 917 series, Olympus AU (400/600/640/680), Beckman Synchron CX, LX and DXC. The reagent is stable for 12 months from date of manufacture when stored @ 2°-8°C. |

 Assay PrincipleThe Diazyme TBA assay is an enzyme cycling based assay utilizing the unique enzymatic property of 3-α hydroxy-steroid dehydrogenase (3-α HSD) that catalyzes the reversible reactions. The substrate bile acids is converted into 3-keto steroids in the forward reaction with concomitant conversion of Thio-NAD to Thio-NADH. In the reverse reaction, 3-keto steroids is converted back to bile acids with concomitant conversion of NADH to NAD. The amount of Thio-NADH formation in the cycling reaction is proportional to the amount of total bile acids in the sample. The formed Thio-NADH is detected at 405nm. |

| Intended use Diazyme Total Bile Acids Assay Kit is intended for the in vitro quantitative determination of total bile acids (TBA) in human serum. Total bile acids are metabolized in the liver and serve as a marker for normal and abnormal liver function. Serum total bile acids are increased in patients with liver disease. |
